12 grown discus in a 50 gal. would seem a bit crowded

Most likely. Depends on size, how much you feed them, biological load i.e how many other fish, plants etc what type of filtration and water turn over per hour. The heavily stocked tanks you see in aquariums have large filtration systems behind the scenes and a clear base.

    Assume one adult discus/40L.

We have 8 Discus in our 550ltr, with a massive Sump, so as stated above, depending on filtration load etc, I personally think its alittle tight

In a word, "YEP". Even if they are only juvies, you need more water. Long term for them to thrive as adults you'll need at least 500 liters. "T"
